Samus Aran is the main character and protagonist of the Metroid series. Introduced to the public in the 1986 game Metroid, Samus is a Human Bounty Hunter famous for completing seemingly impossible missions. Armed with a cybernetic Power Suit built by the Chozo, an ancient intelligent race, she is a powerful opponent in battle. She hunts Space Pirates and energy-absorbing creatures known as metroids, while completing missions given to her by the Galactic Federation. In Metroid Prime, Metroid Prime 2: Echoes and Metroid Prime 3: Corruption she is voiced by Jennifer Hale, but only to indicate damage, groans for minor damage and the scream that indicates death. In Metroid: Other M, Samus is voiced by Ai Kobayashi (Japanese audio) and Jessica Martin (English audio). Samus entered the gaming world in Metroid, which led players to believe that she was a man, since the game's manual gave masculine descriptions of the girl, and she always wore her suits. Thus, people believed that she was a robot or a man inside a robotic armor. However, completing Metroid in less than an hour revealed that Samus was a young and athletic woman. Although she uses the Power Suit in most games, it has become a tradition to show her in more sensual clothing at the end of each game, usually as a reward for meeting certain conditions, such as completing the game quickly or with a high percentage of items collected.

Samus Aran is a native of Earth, born on the colony K-2L. Samus lived with her parents Virginia Aran and Rodney Aran until she was three years old, when the colony was visited by two Chozos. The Chozos contacted the humans hoping to receive some of the Afloraltite that the colony mined, but their request was denied by the inhabitants. Samus, however, caught the attention of Old Bird, one of the Chozo elders. The two went on a hike, where they struck up a friendship. Samus began calling him "Tori Oji-chan" (literally, "Grandpa-bird"), and Old Bird managed to get Pyonchi to become Samus's pet. However, a Space Pirate ship led by Ridley attacked the colony shortly after the Chozos left the planet. During the attack, Samus encountered Ridley; she asked if the two could be friends, repeating the same words as Old Bird, but Ridley responded with an attack that hit Samus and Virginia. Meanwhile, Samus' father, Rodney, sacrificed himself to destroy Ridley's ship, along with part of his pirate crew. Samus was the sole survivor of the attack on K-2L. Some time after the attack, the same group of Chozos who visited the planet later returned, and found the girl accompanied by Pyonchi, and decided to take them to Zebes, since they would not survive alone. However, Samus's constitution was too weak to survive on Zebes for more than a few days, leading to the infusion of Gray Voice's DNA into the child's body, making her adaptable to the planet's hostile environment. Later, at the age of 14, Samus received her Power Suit, built entirely with Chozo technology. Samus was trained for several years by the Chozo, and by the age of 14 she had surpassed the physical limits of any normal human. The Chozo were still hesitant about making her a warrior until they witnessed her compassion for Iona Faerias who had lost their reason and began attacking her.
