Ami Mizuno is an outstanding and highly intelligent student who attends the same school as the series' protagonist, Usagi Tsukino. Usagi, who had already begun to adopt the alternate identity as the warrior "Sailor Moon", helps Ami discover that she possesses a hidden power to also transform into a warrior of justice called "Sailor Mercury". Personality Ami is a shy, introverted and quiet girl who has a hard time making friends and is very beautiful and intelligent; however, she prefers to be alone with her books. Her classmates thought she was arrogant because of her intelligence and misinterpreted behavior, so they did not approach her, until Usagi approached her. She enjoys solitude as she prefers to do things alone than in a group, although after meeting the other Sailor Guardians she gradually develops a taste for meeting up with her new friends. She studies hard every day, and stands out for being intelligent, smart, analytical, and observant, earning the highest grade on national exams, leading her classmates to speculate that she was superhuman and had an IQ of 300. Math and science are her favorite subjects, and she is destined to become a doctor like her mother. She is usually calm and sensible, she is the voice of reason for the team, despite being young she shows a lot of maturity, experience, and seriousness as if she were an older woman, at various times she assumes the role of leader of the group since she is the one who reprimands, advises, and prevents her friends from getting involved in problems, sometimes she assumes a maternal role since she tells the others what to do as if her friends were her daughters. In relation to school, Ami will never let Usagi copy her, as doing so would not teach her anything, just like with Minako. In the first season, Ami and Ryo Urawa showed feelings for each other, but she refused from the start, saying that she had no time for boyfriends. The two eventually went on a date, but Ryo left Tokyo shortly after, and although he appeared in the penultimate episode of the first season as an illusion, he was never mentioned in the series again. In the video game Sailor Moon: Another Story, he returned for vacation, but was attacked by a Yoma who revealed to Ami a prophecy he had seen in a vision, and then disappeared shortly afterward, never to be seen again. In Sailor Moon Sailor Stars, Ami appears to be attracted to Kou Taiki of the Three Lights, who is very intelligent and studious like her. However, Ami's idealism and faith in Sailor Moon is clashed with Taiki's cold reasoning and dislike of her. At one point, when Ami (as Sailor Mercury) finds herself in a bind in a battle against a monster, Taiki leaves her, rather than helping her. They interacted quite a bit during the season, but there was no romance between them. In a picture drawn by Naoko Takeuchi, Ami is cuddling with Zoisite. Occurrences of this pairing are found in some of the musicals. This is because Sailor Mercury and Zoisite were supposed to be a couple during the Silver Millennium.

In the special "Ami-chan no Hatsukoi" based on her, it was revealed that Ami is allergic to love letters and explodes when she receives them. This was briefly mentioned in episode 178. Her Three Lights membership number is 25, the lowest of all her friends, which means she was very excited to be one of the first 30 fans to get it. For some reason, in several dubs the pronunciation of her name is changed from Ami to the English pronunciation of Amy (Eimi), which is actually the diminutive of Amanda.
