The character is based on Professor James Moriarty's lesser-known younger brother, who has been referenced twice in the Doyle canon. As the manga uses the two brothers theory due to Doyle reconfiguring his brother's profession, he is likely based on the Valley of Fear incarnation, whom Holmes referred to as being a "stationmaster".
