Raphael is a mutant alligator snapping turtle and the older brother and leader of the Teenage Mutant Ninja Turtles. In Rise of the Teenage Mutant Ninja Turtles, his weapons are a pair of tonfās, which help him harness the power of smash-jutsu.Personality Raphael has a boisterous and brash personality, preferring to act first and think later when facing enemies. However, he can get anxious under pressure, which is made worse when he becomes shy about it. He has a hard time being mean to people, especially his brothers, which is shown when he didn't want to upset Donatello for not liking his gifts, but will fight with them if they are arguing, such as when he fought with Leonardo over dethroning his wrestling idol Ghost Bear. He seems to have a soft spot for animals, as shown in episode 1 when he saw Mayhem lost and scared at a construction site. He also seems to be an extreme eater, as shown in episode 2 when he ate a large amount of salami (including the ninja salami).

Appearance Raphael is a mutant alligator snapping turtle with bright green skin, an orange plastron with forked upper edges, a dark green carapace with three dorsal ridges, and a large, muscular build. He has spikes on his shoulders and elbows, yellow sclera and teeth, a sharp tooth protruding from the right side of his upper jaw, three fingers on each hand, and two toes on each foot. He wears a red bandana covering the top of his head with long, torn bandage tips. His bandana has a tear under his right eye. He wears red bandages around his elbows, white bandages around his hands and ankles, and red boxer shorts. There are white adhesive bandages over his plastron, left forearm, and right calf, and a red belt with a Turtle emblem affixed to the left. During Rise of the Teenage Mutant Ninja Turtles: The Movie, he gains a hole in the upper left side of his plastron when he takes a hit from Krang Uno in a successful attempt to protect Leonardo.

Curiosities Raphael was named after the famous Italian Renaissance master Raffaello Sanzio da Urbino. The name Raphael/Rafael is of Hebrew origin, meaning "God has healed". Raphael is also the only one of the Ninja Turtles to have a Hebrew name, while the other three Turtle names are Italian. He is the only Turtle to not have an "o" sound at the end of his name. This is because the Ninja Turtles were named after the common English names of four Renaissance artists, and unlike the others, the artist Raphael is not usually referred to in English-speaking regions by his corresponding original Italian name, Raffaello. Unlike previous incarnations of the Turtles, this version of Raphael is both the older brother and the leader of the team at the beginning of the series, rather than Leonardo. If the musical lyrics of the song Turtle Power are anything to go by, this is the second time Raphael has been featured as the leader of the Turtles. Raphael's weapon of choice is a tonfā rather than his usual weapon, the sai, making use of these during the early events of episode 1 before breaking them. This version of Raphael is not the first to wield a tonfā. The earliest depiction of this is in Mirage Studios' 1986 comic TMNT #9, whose story takes place when Mirage's Ninja Turtles are still pre-teens in ninja training, Raphael's primary weapons were tonfās rather than the sais he would later wield. In this iteration, Raphael is an alligator-type snapping turtle, known in real life to be not only the largest species of snapping turtle, but also the largest species of freshwater turtle in North America,[2] hence his large, bulky physique.

In this version, Raphael's physical profile is also similar to that of several incarnations of Slash, most of which are also snapping turtles. He also resembles Raphael from the 2014 film series.

According to Leonardo, Raphael is also considered a "figure it out as you go" type of leader. Describing his leadership role in a live broadcast, Raphael was described as the "reactionary type leader." As described in a tweet by Russ Carneyel, writer for the animated series, "Raph is all heart, all loyalty, all crushing, but maybe not so strong on tactics?"[4] Donatello reveals that Raphael is allergic to peanuts (episode: "Mascot Melee"). Raphael's surprised exclamation, "Jumpin' Jack Flash!", may be a reference to the Rolling Stones song of the same name or the 1986 film of the same name. A recurring gag is that when Raphael is talking on the phone with his brothers, he speaks in a soft, unintentionally condescending tone of voice. Raphael apparently has a social media account that he posts on (episode: "Mind Meld"). Raphael's favorite music genre is R&B (episode: "Al Be Back").[5][6][7] Despite Raphael's affection for animals, they tend to fear or hate him, probably due to his gigantic stature. Raphael wears a retention headset with his pajamas (episode: "Flushed, but Never Forgotten"). Raphael claims he can fit his entire fist in his mouth (episode: "Lair Games"). As stated by Donatello, Raphael gets stuck using a pirate voice every time he watches Jupiter Jim Sails the Seven Galaxies (episode: "Snow Day"). Raphael has a habit of impulsively pressing big red buttons (episode: "Cloak and Swaggart"). According to Leonardo, Raphael is "terrible at taking photographs" (episode: "Mind Meld"). It is not unusual for Raphael to get into physical fights with himself. He is shown doing this several times throughout the series (episode: "Pizza Puffs"). Raphael confesses that he has a "phobia of tanks" (episode: "Cloak and Swaggart").
